Abstract

 Background & Objectives: Spironolactone is a diuretic and antiandrogenic drug and is used in the treatment of hypertention 'secondary hyprealdosteronism congestive heart failure' cirrhosis of the liver, nephrotic syndrome 'androgenic alopecia' gynecomastia and hirsutism. In this research, the effects of spironolactone on the serum LH, FSH, testosterone, dihydrotestosterone, and changes in body weight and testicular tissue in adult male rats, were studied.

 Methods: For this purpose 190 10 g male wistar rats (n=40) were randomly divided into the following grups: control, sham operated (received water) and 25, 50, 100 mg/kg oral spironolactone treated groups. After 14 days body weight and testis weight under laboratory methods, were measured and blood samples were taken from heart and used for the measuring of LH/FSH/testosterone and dihydrotestosterone and then the rates' testes, in order to evaluate the histological changes, were removed and weighed and after obtaining tissue section and staining through HE, they were studied.

 Results: Serum LH level showed a significant increase and testosterone and dihydrotestosterone levels showed asignigicant decrease in 100mg/kg spironolactone treated group ( p 0.05 ) and there was no significant difference among serum FSH level, body weight and testicular weight as compared to control group.

 Conclusion: It can be concluded that oral administration of spironolactone maximum dose for 14 days could increase serum LHlevel and decrease testosterone and dihydrotestosterone levels.

Abstract

Background & Objective: Many factors interfere in liver fibrosis one of which is hepatitis B.The aim of our study was to determine the parameters serum iron, TIBC and ferritin of patients with hepatitis B and fibrosis of the liver

Methods: Study involved 50 cases with liver fibrosis and viral hepatitis type B (2002-2004)
and 26 controls. Iron concentration, Total iron-binding capacity and Ferritin concentration were measured by standard biochemical methods. To compare the results of the two groyps a student t-test was used and considering p< 0.05 the difference between two groups was significant.

Results: The results showed that serum ferritin increase 243.7±16.7 micro gr/L and 238.7±13.9 micro gr/L in man and women patients with chronic hepatitis B virus infection compared to the control group, 198.5±14.3 and 182.7 ±14.3 mg/L in man and women, respectively. No significant difference was found between serum Iron and TIBC in both groups. there was a significant relationship between them.

Conclusion: Increased serum ferritin concentration and liver fibrosis induced by hepatitis B virus infection. Also, determination of serum ferritin could help in diagnosis of liver fibrosis.

Abstract

 Background & Objective: During pregnancy, Hb and Hct are reduced, because of increase in maternal plasma volume. Pregnant women need more iron, because of the increase in blood volume, blood cells, the growth of fetus and placenta, iron supplementation is performed. This study, measured blood index after different doses of iron supplement prescribed in healthy pregnant women in order to prevent excessive iron intake.

 Methods: In this cross-sectional analytical study, 150 healthy pregnant women were selected who were divided in to two groups. One group took daily (as control) and the other intermittent (as case=3 times in week) iron supplement from the second trimester. Blood samples of all cases in the first second an third trimester were analysed for complete blood counts (CBC). The Data were analysed by SPSS.

 Results: The reduction of Hb, HcT and RBC at second trimester was significant compared to the first (p<0.05). This difference was not significant among two groups concerning the blood index.

 Conclusion: Since there was no significant difference in blood index among the case and control groups, so, to prevent the side effects of iron over load, it’s recommended, to use intermittent iron supplement in healthy pregnant women.

Abstract

  Background & Objectives: The development of technology has naturally given rise to an increase in environmental low-frequency electromagnetic fields and consequently has attracted scholars' attention. Most of the studies have focused on transmission lines and power system distribution with 50 Hz. This research is an attempt to show the effect of 50 Hz magnetic fields on bioelectric parameters and indicates the possible influence of this change in F1 cells of Helix aspersa .

  Methods: The present research used Helix aspersa neuron F1 to identify the location of magnetic fields as well as the rate of effects of environmental magnetic fields on nervous system. Control group was used to study the effect of elapsed time, electrode entering and the cell membrane rupture. Intuition group and environmental group were considered in order to study the potential impact of interfering environmental factors and identify the effectiveness rate of magnetic fields, respectively. For the purpose of producing uniform magnetic field Helmholtz coil was used. Electrophysiological recording was realized under the requirements of current clamp. And, in order to show the impacts from magnetic fields on ion channels Hodgkin-Huxley cell model was applied. All data were analyzed taking the advantage of SPSS 16 software and two-way ANOVA statistical test. P < 0.05 was considered as significance level. And MATLAB software environment and PSO were used in order for applying the algorithm and estimating the parameters.

  Result: No statistically significant difference was found between control and sham groups in different time intervals. Once the 45.87 microtesla was applied significant differences were observed 12 minutes after the application. The highest amount of change happened 14 minutes after the application of more fields. With the application of the field, the amplitude of the sodium action potential shows decreasing trend . No significant changes were observed in different time intervals, whereas significant differences were seen in frequency of action potential during different time intervals. The amplitude of AHP shows no significant changes .

  Conclusion: The results indicated that low-frequency magnetic fields with 50 Hz frequency will directly lead to change in bioelectric activities of neurons through a change in amount and rate of opening and closing of ionic channels and the conductivity of sodium and potassium channels reduces together with increase in conductance of potassium dependent calcium channel (AHP).

Abstract

Background & objectives: Salvia officinalis is one of the memory-enhancing herbs that were used in the past. On the other hand, iron oxide nanoparticles which are widely used in medicine and industry may impair the brain process related to memory. In this investigation, the effect of hydro-alcoholic extract of Salvia officinalis on iron oxide nanoparticle induced memory impairment and the role of beta-adrenergic receptors in this effect were studied.

Methods: To assess the inhibitory avoidance memory, animals were trained in the step-down task and drugs (saline, hydro-alcoholic extract of sage leaves, nanoparticles of iron oxide and propranolol) were injected immediately after training by intraperitoneal (ip) injections. Long-term memory was tested 24 hours later and step-down latencies were recorded.

Results: Administration of iron oxide nanoparticles (5 mg/kg, ip) impaired memory retrieval. Salvia officinalis extract (40 mg/kg, ip) also prevented iron oxide nanoparticle induced long-term memory impairment. On the other hand, administration of propranolol (5, 10 mg/kg, ip) before Salvia officinalis extract (40 mg/kg, ip) and iron oxide nanoparticles (5 mg/kg, ip) attenuated the effect of Salvia officinalis extract.

Conclusion: It seems that extract of Salvia officinalis leaves decreases iron oxide nanoparticle induced memory impairment. Beta-adrenergic mechanisms are possibly involved in these effects of Salvia officinalis extract.

Abstract

Background & objectives: Iron overload is involved in the pathophysiology of many diseases including diabetes. In fact, the excess iron by creating free radicals makes damage to pancreas and leads to insulin resistance and diabetes. Andrographolide extract has hypoglycemic and antioxidant properties. This study has surveyed the effects of andrographolide on blood glucose and lipid profile in rats with secondary iron overload.

Methods: In this experimental study, 36 male Wistar rats were randomly divided into 6 groups: the healthy control group, secondary iron overload group, secondary iron overload groups treated with a dose of 3.5 and 7 mg/kg of andrographolide extract, and andrographolide groups treated with a dose of 3.5 and 7 mg/kg of extract. Iron and extract were injected for 6 and 12 days, respectively. Blood samples were taken for measurement of blood glucose and lipid profiles. Data were analyzed using ANOVA test.

Results: The pathological results of samples from liver of animals receiving iron showed that the iron was deposited in the liver tissues. Iron injection significantly increased blood glucose levels compared to healthy control group (p<0.05). In the iron overload group, andrographolide extract with a dose of 3.5 mg/kg or 7 mg/kg significantly decreased blood glucose levels (p<0.05). Iron injections did not increase the serum triglyceride and cholesterollevels. Injections of andrographolide extract with a dose of 3.5 mg/kg and 7 mg/kg, significantly decreased the cholesterol levels compared to iron receiving group (p<0.05).

Conclusion: Results of this study showed that the andrographolide with different doses may be effective in the treatment of diabetes by reducing serum glucose and cholesterol levels.

Abstract

Background & objectives: Andrographolide has potent antioxidant properties. There is still not a complete understanding of the antioxidant effects of andrographolide. In this study, the effect of andrographolide on the serum antioxidant activity of superoxide dismutase and catalase was investigated in the rats with secondary iron overload. 
Methods: In this experimental study, 32 rats were divided into 4 groups of 8 each. The groups included: 1-Control 2- Receiving iron 3- Receiving iron treated with 7 mg/kg andrographolide 4- Receiving 7 mg/kg andrographolide. The serum activity of superoxide dismutase and catalase and serum level of malondialdehyde (MDA) was measured using colorimetric method. Prussian blue was used to evaluate iron deposition in liver.
Results: The results showed that iron was deposited in the liver. The activity of superoxide dismutase and catalase in the secondary iron receiving group, compared to control group, was significantly increased (p<0.05). The activity of catalase was significantly decreased in the group receiving iron and treated with andrographolide, compared to the control and iron receiving groups (p<0.05). Also, the activity of superoxide dismutase in the group receiving iron and treated with andrographolide decreased significantly compared to the iron receiving group (p<0.05). Serum levels of malondialdehyde in the iron receiving group were higher than the control group (p<0.05). Serum levels of malondialdehyde in the group receiving iron and treated with andrographolide, compared with the iron receiving group, showed a significant decrease (p<0.05).
Conclusion: Andrographolide (7mg/kg) reduced the serum activity of catalase and superoxide dismutase and malondialdehyde serum levels in rats with secondary iron overload.

Background & objectives: So far, various reports have been presented on the relationship between sex hormones and gender-related differences in pain and analgesia in humans and laboratory animals. The purpose of this study was to investigate the effect of testosterone hormone and spironolactone anti-androgen drug on morphine-induced analgesia in male mice using formalin test.
Methods: In this study, 80 male mice were divided into 10 groups (N=8); normal saline (control), sesame seed oil (as testosterone solvent), testosterone (5 and 10 mg/kg body weight), spironolactone, morphine, sesame seed oil + morphine, testosterone (5 and 10 mg/ kg body weight) + morphine and spironolactone + morphine. Formalin test was performed in all the mice, and data were analyzed by one-way ANOVA.
Results: The results showed that sesame seed oil + morphine (p<0.001), morphine (p<0.001), testosterone (5 mg/kg) + morphine (p<0.01) and testosterone (10 mg/kg) + morphine (p<0.001) significantly reduced acute  pain, and testosterone (5 mg/kg) (p<0.05), testosterone (10 mg/kg) (p<0.01), sesame seed oil + morphine (p<0.001), morphine (p<0.001), testosterone (5 mg/kg) + morphine (p<0.001) and testosterone (10 mg/kg) + morphine (p<0.001) significantly reduced chronic pain compared with control group. Spironolactone had no effect on pain relief in the presence and absence of morphine compared to control group.
Conclusions: It can be concluded that testosterone has analgesic effects on the chronic phase of the pain. On the other hand, spironolactone may have hyperalgesic effects due to its anti-androgenic properties.

Abstract

Background & objectives: Caffeine can affect many physiological functions of the body. The aim of this study was to investigate the response of aerobic function, blood pressure and some immune system factors of trained men to acute caffeine consumption in dehydrated condition and warm and humid environment.
Methods: Thirty male athletes with a mean age of 26.6±3.9 years in dehydrated conditions voluntarily participated in this study, which was performed in a double-blind manner. Subjects were divided into three groups: caffeine consumption group (n=10), placebo group (n=10) and control group (n=10). The Caffeine group consumed 6 mg/kg body weight of caffeine. Sixty minutes later, the subjects performed an increasingly exhausting exercise. Blood samples collecting and blood pressure measuring took place before, immediately after and 24 hours after exercise. Analysis of variance with repeated measures (group * time) and one-way analysis of variance were used for statistical calculations.
Results: The duration of the exhaustion test in the caffeine consumption group was significantly longer than the control (p≤0.01) and placebo (p≤0.05) groups. In the blood sampling immediately after exercise, the level of leukocytes, neutrophils and lymphocytes in the exercise groups were significantly higher than the control group (p≤0.001). Also, the caffeine group experienced a lower increase in leukocytes and neutrophils compared to the placebo group (p≤0.001). Blood pressure values ​​in the exercise groups were significantly higher than the control group (p≤0.001). Also, the caffeine group experienced a greater increase in systolic blood pressure at baseline and immediately after exercise compared to the placebo group (p≤0.001).
Conclusion: In general, exhausting exercise in a warm environment and dehydrated condition increases the number of immune system cells and blood pressure. In addition to improving aerobic function, caffeine seems to prevent further increases in the number of immune system cells, while increasing blood pressure in the normal range in warm and humid environment.
