Abstract

  Background & objectives: During lifetime, hair follicles undergo three cyclic changes: anagen, catagen, telogen. In hair follicles, stem cells located in Bulge area, which is part of the outer root sheath. Bulge cells proliferate new cells in anagen phase. Bulge region of hair follicle indicated as a source of stem cell for many years, but little studies done in vitro to characterize rat bulge region hair follicle stem cells.

  Methods: In this study Bulge cells of rat hair follicle were isolated and cultured, then morphological features of these cells surveyed. Nestin and CD34 as hair follicle stem cell markers, and K15 as a keratinocyte marker assessed by immunocytochemistry after one to three weeks.

  Results: In this study, we found that, 2 days after attachment of cells to floor of plates, the cells were initiated to proliferation and migration. These cells had good nestin and CD34 immunostaining, but after three week differentiation,were nestin and CD34 negative. Also these cells couldn’t express K15.

  Conclusion : Results showed that cultivated rat bulge cells, have high proliferative potential, and also could express nestin and CD34 as stem cell factors.

Abstract

  Background & Objectives: Using of drugs arbitrarily leads to failure of treatment and causes side effects in prolonged time. The aim of this study was to survey prevalence of arbitrary drug use and the factors affecting it.

  Methods: Three hundred students participated in this cross-sectional and descriptive study in Ardabil (Islamic Azad, Mohhaghegh Ardabili, Payame-Noor and Medical Sciences Universities) during 2010. The samples were selected randomly from four universities and data were collected by questionnaire and then, analyzed by descriptive and analytical statistics such as chi-square test.

  Results: The average of students’ age was 20.64±2 years. 23.3% of them were married from whom 92.2% were non-medical students and 7.1% were medical students. 76.7% of all students were single. 83% of all students were using drugs arbitrarily. The highest mean score for student attitude was reflected to safety of used drugs. Lack of time to visit a physician, believing the safety of many drugs and selling of drugs without prescription were the most common causes in arbitrary drug use among students.

  Conclusion: Regarding to the high arbitrary drug use among students, it is necessary to make a new policy for education and correct informing of students in order to reduce self-treatment.

Abstract

  Background &Objectives: Stem cells are a class of undifferentiated cells that are able to differentiate into specialized cell types. The discovery of such cells in the adult mammalian central nervous system (CNS), an organ traditionally thought to have little or no regenerative capacity, opened the door to treatment of degenerative diseases of CNS like Stroke, Parkinson, Alzheimer and Spinal Cord Injury. Thus, here we described the isolation of neural stem cells from the adult mouse brain using the neurosphere assay (NSA) and differentiation of these cells to neural adult cells in details.

  Methods: The rostral part of the subventricular zone (SVZ) of the lateral ventricles in the adult mice was dissociated into single cell suspension and cultured using NSA. Primary neurospheres were counted seven days after plating and then the mean number of neurospheres was recorded. The differentiation of neural stem cells into adult neural cells was accomplished by plating the neurosphere-derived cells in differentiating media. Immunocytochemistry and specific markers were used for the identification of the adult neural stem cells.

  Results : The cell suspension obtained from the rostral part of the SVZ of the lateral ventricles generated multipotential colonies, called neurospheres, 7 to 10 days post- incubation. The mean number of neurospheres generated from SVZ was 505±62. The multipotentiality of the neurospheres was shown by palting them in differentiating media and generating adult neural cells including neuron, astrocyte and oligodendrocyte .

  Conclusion: Owing to their rarity and paucity of neural stem cell specific markers, the NSA is a common and selective method for isolating and understanding the biology of embryonic and adult neural stem cells.

Abstract

  Background & Objectives : Several studies have been conducted to explore anti-HIV drugs. Discovery and study of novel anti-HIV-1 compounds need live viruses and has serious biosafety concerns. In this research we reported a novel and safe system for assaying the cytopathic effects of HIV by using single cycle replicable (SCR) HIV-1 virions.

  Methods: To produce the SCR HIV-1 virions, pMD2G, pmzNL4-3 and pSPAX2 plasmids were co-transfected into HEK293T cells. Different amount of SCR virions were used to infect target cells (MT-2). Within the infected cells, the number of formed syncytia was counted under the light microscopy. The lethal effects of the SCR HIV virions were measured using XTT proliferation assay.

  Results: Formation of syncytia among SCR HIV infected cells was detectable 24 hours after infection. Highest amount of syncytia was seen 72 hours after infection. Increase in the amount of virions caused increasing of syncytia and the cytopathic effects of SCR HIV-1. Infection with more than 1600ng P24 SCR HIV decreased the syncytium formation and viability of all cells. The calculated IC50 (50 percent inhibitory capacity) for nevirapine and BMS806 using this method was 50nM and 30nM, respectively.

  Conclusion: SCR HIV-1 virions are replicable only for one cycle. Using these virions can improve the safety of HIV researches. Herein, we optimized the assaying of HIV induced cytopathic effects by using SCR HIV-1 (NL4-3) virions. The accuracy of this method was accepted by quantifying the anti-HIV-1 effects of nevirapine and BMS806 by (SCR) HIV-1 virions.

Abstract

  Background & Objectives: Coronary artery disease is the main cause of mortality in developing and industrial countries. Recently the involvement of infectious agents as a risk factor for Acute Coronary syndrome is drafted. So this study was designed to investigate the probable association between Acute Coronary syndrome and Helicobacter pylori infection.

  Methods: This case-control study was carried out on 300 hospitalized patients with the diagnosis of Acute Coronary syndrome (UA and MI) and 300 hospitalized patients without the history of coronary heart disease. Anti Helicobacter pylori Antibody level was determined by as an indicator of infection history. Using chi-square and t- test the results were analyzed in SPSS software.

  Results: Results showed that 79 patients (26.3%) in control group and 122 patients (40.6%) in case group were seropositive and the difference was significant. Relationship between cronory diseases risk factors and levels of IgG was not significant. Also the results showed that the rate of hypertension in seropositive patients in case group was significantly upper than control group.

  Conclusion: Regarding the findings of this study we can conclude that Helicobacter pylori infection probably is a risk factor for Acute Coronary Syndrome. Thus, further studies are needed to elucidate the association between Helicobacter pylori infection and Acute Coronary Syndrome.

Abstract

  Background & Objectives: Polycystic ovary syndrome (PCOS) is one of the major endocrine disorders among females. PCOS is associated with development of type 2 diabetes and cardiovascular disease. The objectives of this study were to determine the effects of omega-3 fatty acids on glycemic status and high sensitive C-reactive protein in women with PCOS.

  Methods: This double-blind randomized controlled clinical trial was conducted on 61 PCOS patients. Subjects in ω-3 fatty acids (n=30) and placebo (n=31) groups take 4 ω-3 fatty acids capsules (each one contained 180 mg eicosapentaenoic acid and 120 mg docosahexanoic acid) or placebo daily for 8 weeks. Anthropometric and biochemical measurments and food intakes were assessed at the beginning and at the end of the study.

  Results: ω-3 fatty acid supplementation caused significant decrease in serum levels of glucose, insulin (p < 0.001 for both) and insulin resistance (p = 0.002) at the end of the study in comparision with baseline values. This was also statistically significant compared to the placebo group (p < 0.001, p = 0.002 and p = 0.001, respectively). No statistically significant differences in serum levels of high sensitive C-reactive protein were obsereved in any of groups at the end of the study (p > 0.05).

  Conclusion: ω-3 fatty acid supplementation had some beneficial effects on glycemic status of PCOS patients and may be useful in prevention and control of metabolic conmplication of this syndrome.

Abstract

  Background & Objectives: Type 2 diabetes is a major cardiovascular risk factor such as HTN, HLP and smoking. A primary diabetic cardiomyopathy represents a high risk factor for heart failure in the absence of ischemic, valvular and hypertensive heart disease in the diabetic population. CAD is more common in diabetic patients and it is almost asymptomatic.

  Unquestionably, an early detection of LV damage and CAD is a major goal for the prevention of cardiac disease in the diabetic population.

  Methods: This study was done as Cross-Sectional method. The study sample consisted of 40 patients with type 2 diabetes mellitus without hypertension and cardiac symptoms (mean age 47 years) who recourse to diabetes clinic of Ardabil Imam Khomeini Hospital during 2009-2010. Left ventricular (LV) function was studied by echocardiography and exercise test using Bruce protocol. Data from the patients were collected and analyzed using SPSS 17 software.

  Results: All studied cases had a normal systolic function. 22 cases (55%) had diastolic dysfunction and 8 people (20%) had a positive stress test, which all had diastolic dysfunction too.

  Conclusion: This study showed that an impairment of left ventricular diastolic function occurs early in the natural history of diabetes mellitus and CAD is more common in diabetic patients with diastolic dysfunction.

Abstract

  Background & Objectives : Visceral leishmaniasis is an important endemic disease in Iran and Meshkin shahr County is the most important foci of visceral leishmaniasis in Iran. The aim of this study is the survey of recent changes in epidemiological patterns of visceral leishmaniasis in Meshkin Shar area during 12 years.

  Methods : This is a descriptive retrospective study. Records of 193 cases with confirmed kala-azar disease were reviewed in the period of 2001-2012. Data collected from files and were analyzed using descriptive statistics and SPSS v.15.

  Results : From 193 patients with confirmed kala-azar disease, 85% were resident in rural and 15% in urban areas. Among of these patients, 43% were female and 57% male. About 66% of patients were under 2 years old, 23% in age group between 2-5 years and 11% above five years. Among four parts of Meshkin Shahr County, in the first five years of the study, the most cases of the disease were from the central part, but in second six years the most cases were from the west part of this county and in latest year (2012) it has been coverted .

  Conclusions : Incidence of kala-azar was decreased during last 12 years. Because of high infection rate in dogs and considering important role of these reservoirs in conserving and distribution of disease, stopped health activities in any parts of area can increase the incidence of disease.

Abstract

  Background & objectives: Heart failure is considered as a major cause of hospitalization. Many studies have shown association between sleep-related breathing disorders and heart failure. It has been shown that the relationship between nocturnal hypoxia and left ventricular dysfunction can cause significant morbidity and mortality in patients with congestive heart failure (CHF ). Accordingly, treatment of sleep related breathing disorders (SRBD) can give rise to improvement in CHF treatment too. This study surveys the prevalence of sleep disorder in stable heart failure patients regardless of ejection fraction.

  Methods : This study was a descriptive-analytical study. One hundred and eight patients with heart failure disease were studied. A questionnaire consisting of two parts (part I consistent of demographic information and part II consistent of sleep disorders) and clinical examination (pulse oximetery and echocardiography) were used for collection of data. The data were analyzed with SPSS statistical software using descriptive and analytical tests including the chi-square, Pearson correlation and ANOVA.

  Results: Fifty six persons (51.9%) of patients were female and 52 persons (48.1%) were male with mean age of 65.42 ± 11 years. In total sleep duration, 95 patients (88%) had nocturnal hypoxemia . There was correlation between arterial oxygen desaturation at night and ejection fraction .

  Conclusion : This study confirmed strong associations between nocturnal hypoxia and left ventricular dysfunction and SRBD should be considered in clinical treatment of systolic heart failure.

Abstract

  Background & objectives: Addiction is one of the problems in world threating the social, economic and culture factors. It is essential to have an accurate knowledge about the characteristics of drug users in order to diminish the high-risk behaviors of intravenous drug addicts. This research has been done to assess relationship between individual characteristics and high risk behavior in intravenous drug addicts.

  Method: In this descriptive-analytic research, 360 drug users were selected from different places in Ardabil city and interviewed by a prepared questionnaire. The data were analyzed using descriptive and analytical tests including t-test, Pearson correlation and ANOVA with SPSS statistical software.

  Results: The results showed that the age, gender, material status, job position, age of addiction start, age of injection start, injection frequency, injection frequency per day, syringe supply place and the partner’s gender during recent few months had no significant difference compared to drug users with needle sharing and without needle sharing. The educational level of drug users with needle sharing was lower (P=0.037) and the number of new syringe usage per month was also lesser (P=0.001). They predicted to be more likely infected with AIDS (P=0.001) and had a less argument with their partner about using condom, also mostly have not used condom at their last sexual relationship (P=0.001). The average number of their partners during last three months was high (P=0.003) and there was a meaningful relationship between true sense of peril and using condom in drug users with needle sharing group (p=0.001).

  Conclusion: There is a significant relationship between the true sense of danger and the using condom. It is necessary to have an appropriate advertising to increase using condoms among injecting drug users.

Abstract

  Background & objectives: Procalcitonin (PCT) is a precursor of calcitonin hormone, comprising 116 amino acids, which is produced in thyroid neuroendocrine cells. However, in conditions such as septic shock, metastatic cancers, bacterial and fungal infections PCT is secreted by lung neuroendocrine cells, liver cells, and leukocytes. PCT is involved in acute phase of inflammatory process as a reactant protein. Hence, in this study serum and pleural fluid PCT was evaluated in common types of pleural effusion.

  Methods: This is a cross-sectional case-series study carried out on 60 patients with pleural effusion. In this study, effusion cases due to tuberculosis, malignancy, effusion followed by pneumonia, and also transudate were investigated. Other cases of pleural effusion were excluded from this study. After collecting the samples, PCT levels in pleural fluid and serum of patients were measured and compared statistically with respect to the primary etiology of effusion.

  Results: Sixty patients (10 patients with TB, 10 patients with malignancy, 10 patients with para-pneumonic, and 30 patients with transudative pleural effusion) participated in this study in which 61.7% were men and 38.3% women (p=0.204). The mean age of participants was 62.68 ±19.38 years. Results showed that mean of PCT in pleural fluid of patients with exudative effusion was 0.74 ng/ml (0.63 ng/ml in malignancy, 0.42 ng/ml in TB and 1.51 ng/ml in para-pneumonic) and in patients with transudative was 0.42 ng/ml. In addition, mean of PCT in the serum of patients with exudative pleural effusion was 0.74 ng/ml (0.65 ng/ml in malignancy, 0.40 ng/ml in TB and 1.20 ng/ml in para-pneumonic) and in patients with transudative was 0.43 ng/ml. The analyzed data showed that there was a significant difference in the pleural fluid (p=0.016) and serum PCT (p=0.009) of patients with pleural effusion transudate and exudates. After excluding patients with transudative, a significant difference was also observed among patients with exudative in such a way that in the para-pneumonic group the amount of PCT level in the serum and pleural fluid was more than tuberculosis and malignancy groups.

  Conclusion: Serum and pleural fluid PCT is increased as a marker in response to inflammation. Although its increase in patients with bacterial and pneumonia infection is significant, it can be increased in patients with secondary inflammation of tuberculosis or malignancy and consequently can be used as an inflammatory indicator in the evaluation of patients with pleural effusion.

Abstract

  Background & objectives: Trichomonas vaginalis is a protozoan parasite lives and also causes symptoms in urogenital system of human. Trichomoniasis is the widest spread infection which is transmitted through sexual contact and more than 170 million people are a ffected with Trichomonas vaginalis in the world. Trichomonas vaginalis in women causes pelvic inflammatory disease, increase in the risk of fallopian tube dependent infertility, ectopic pregnancy, preterm labor, the birth of low weight infants and the increase in the possibility of HIV transmission. Considering the importance of Trichomonas vaginalis infection in pregnant women and lack of data from Ardabil city, this study was performed to identify this infection in pregnant women referred to health and medical centers of Ardabil city.

  Methods: Vaginal discharge from 500 pregnant women was collected with sterile swap and disposable speculum and examined for Trichomonas vaginalis by direct microscopic examination and cultured in Diamond specific medium. A testimonial and questionnaire were completed for each case and the results analyzed using chi-square test by SPSS statistical software version 19.

  Results: In this study, the culture of samples displayed 12 positive cases (2.4%). Furthermore, Trichomonas vaginalis trophozoites were observed in five cases (1%) with direct microscopic examination. Among 12 positive cases, five respondents (41.7%) were in age range of 16-25 years and seven (58.3%) in 26-35 years old. Among different clinical manifestations there was a significant relation between discharge and the infection.

  Conclusion: The present survey confirmed the presence of Trichomonas vaginalis infection in pregnant women of Ardabil city. Therefore, an effective healthcare program by health authorities for prevention of infection in this group seems to be needed.

Abstract

Background & objectives: Recurrent spontaneous abortion (RSA) is defined by two or more consecutive miscarriages before 20 weeks of gestation. Adenosine deaminase (ADA) is an enzyme of purine salvage pathway and has two important isoenzymes ADA1 and ADA2. The adenosine deaminase G22A polymorphism (ADA*2) increases the level of adenosine. Adenosine may play a protective role against recurrent spontaneous abortions, since it regulates blood flow into the uterus and placenta. In consideration of the effect of decreased enzymatic activity of adenosine deaminase G22A polymorphism on adenosine levels we evaluated the protective effect of ADA*2 allele against recurrent spontaneous abortions in north-west of Iran. 

Methods: A total of 100 women were recruited to form two groups. First one, with a history of recurrent spontaneous abortions (N=50), and the second one, without a history of abortions (N=50). Genomic DNA was extracted from peripheral blood with a commercial kit and PCR-RFLP analysis was used to identify the G22A genetic polymorphism. Fisher's exact test and odds ratio values were used to compare the proportions of adenosine deaminase genotypes and alleles between women with and without a history of recurrent spontaneous abortion (p<0.05). 
Results: The frequency of homozygotes (AA) was 2% in control group, whereas no homozygote (AA) was found in the case group. The frequency of heterozygotes (AG) was 20% in control group and 8% in the case group (p<0.05). The frequency of homozygotes (GG) was 78% in control group and 92% in the case group (p<0. 05). A significant increase in the frequency of AG genotype in controls (p=0.014, OR=0.348) relative to women with the history of RSA demonstrates the protective effect of AG genotype in controls.
Conclusion: The data suggest that women carrying the G22A polymorphism (ADA*2 allele) and AG genotype which is associated with the lower enzymatic activity are better protected against recurrent spontaneous abortions.

Abstract

  Background & objectives: Cancer is one of the most causes of mortality in worldwide. Components derived from natural plants that induce apoptosis are used for cancer treatment. Therefore investigation of different herbal components for new anti-cancer drug is one of the main research activities throughout the world. According to low cost, oral consumption and easy access to the public extracts of Urtica dioica, in this study we aimed to investigate the effectiveness of this herb on MDA-MB-468 breast cancer cells.

  Methods: Cytotoxic effect of Urtica dioica extract was measured using MTT assays. To show induction of apoptosis by this plant TUNEL and DNA Fragmentation test were performed.

  Results: In the present study dichloromethane extracts noticeably killed cancer cells. IC50 values related to human breast adenocarcinoma cell line MDA-MB-468 were 29.46±1.05 µg/ml in 24 hours and 15.54±1.04 µg/ml in 48 hours. TUNEL test and DNA Fragmentation assay showed apoptotic characteristic in the extract treated cells.

  Conclusion: The results showed that MDA-MB-468 cells after treatment with dichloromethane extract of Urtica dioica, induces apoptosis in MDA-MB-468 cancer cells which may be useful in the treatment of cancer.

Abstract

Background &objectives: Celiac disease is a gastrointestinal disorder that genetic factors have a role on its pathogenesis. The aim of the present study was to determine HLA alleles encoding HLA DQ2 and HLA DQ8 in patients with celiac disease.

Methods: This descriptive analytical study was conducted on 105 patients with IBS whom referred to the gastroenterology unit in Ardabil, Iran. The patients with positive celiac serology of IgA anti-tTG test were done an endoscopic duodenal biopsy. HLA DQ2 and DQ8 testings were performed on patients. Data were analyzed using descriptive statistics together with Mann-Whitney U and Fisher&rsquos exact tests by SPSS-16.

Results: 14 patients were found positive for IgA antibodies against tTG. From these 14 patients 2 (14.3%) were male and 12 (85.71%) female. All of 14 patients (100% of case) had also abnormal intestinal pathology according to the modified Marsh classification and 10 patients had type IIIC. 11 patients had only HLA DQ2, 2 patients had only HLADQ8, and one patient had both HLADQ2 andDQ8.

Conclusion: Most of patients had positive HLA D Q2. The present study emphasizes that HLA genotypes are an important background to CD development.

Abstract

Background & objectives: Stroke is third leading cause of death and disability in the most of human communities. The use of herbs and medicinal plants in different countries is increasing. Today, herbal medicine is used as alternative or complementary therapies with a fewer side effects. Nigella sativa has a rich medical and religious history. Oxidative stress has important role in the pathophysiology of stroke. As Nigella sativa has antioxidant effects, its administration may produce a protective effect against complications of this disease. We examined the effects of the treatment with Nigella sativa oil on the cerebral infarction and edema.

Methods: 48 Male Sprague-Dawley rats were divided into three groups, sham, control ischemic and Nigella sativa oil treated (2 ml/kg) ischemic groups. Transient focal cerebral ischemia was induced by 90-min-long occlusion of the left middle cerebral artery followed by 24-h-long reperfusion. Neurological deficit score was evaluated at the end of the reperfusion period. Thereafter, the animals were randomly selected and used for two projects: (i) Measurement of the infarct volumes and neurological outcome (ii) investigation of ischemic brain edema formation using a wet/dry method.

Results: Induction of cerebral ischemia in the control group produced considerable brain infarction in conjunction with impaired motor functions and severely brain edema. Treatment with Nigella sativa oil significantly reduced the infarct volume and improved the motor functions. The water content in the left (lesioned) hemisphere was considerably elevated in the control ischemic group. Administration of the Nigella sativa oil significantly lowered the water content in the ischemic lesioned hemisphere.

Conclusion: Treatment with Nigella sativa oil can noticeably decrease the ischemic brain injury, attenuate edema formation and improve motor disabilities.

Abstract

Background & objectives: One of the most proven trajectories for analyzing the drug consumption patterns in the community is to assess the prescriptions. Using the antibiotics for prevention and treatment of infectious diseases not only leads to the loss of pathogens but also causes various complications, the most important of which is the drug resistance. The aim of current study was investigating the different antibiotics prescript by general practitioners (GPs) for outpatients in Ardabil city.

Methods: This study is designed based on the cross-sectional method. The number of 2,000 copies (according to Morgan) from total of 526000 copies sent to Ardabil health insurance during the 4 seasons of 2013 were randomly evaluated. The collected data were analyzed by descriptive statistics, ANOVA and T-Test using spss software. The P ≤0.05 was considered statistically significant.

Results: In this study the average drug per prescription was 3.6 and 54.9% of the prescriptions included antibiotics so that the cefixime, azithromycin, Co-amoxiclave and penicillin 6.3.3 were the most frequently prescribed antibiotics, respectively. The highest prescriptions belonged to penicillins (39.4%) cephalosporins (27.7%) and macrolides (18.3%). The most expensive antibiotic was cefixime tablet (32% of the total cost). The most of the antibiotics were prescribed to be taken orally (tablets or capsules). The mean price of each prescription was 74539±71050 Rials. The highest rate of antibiotic prescription was related to the autumn and then winter and there was a significant relationship between the the number of antibiotics in different seasons of the year (p = 0.005).

Conclusion: The average drugs of each prescription are in accordance with local statistics, but much higher than the global figures. The frequency of antibiotics prescription was higher than the global, several Middle Eastern and even African countries’ indices but was lower than the average level of some regions in the country. The pattern of prescribing antibiotics among the Ardabil GPs shows the more drugs in number and expensive broad-spectrum. The average price of prescripts was higher than the national average. These findings suggest that prescription and using antibiotics is not desirable in Ardabil.

Abstract

Background & objectives: Stroke is third leading cause of death and disability in the most of human communities. Several experimental studies have shown that combination therapy with drugs that act via different mechanisms can produce amplified protective effects. We examined the effects of combination therapy with candesartan and alpha tocopherol against cerebral ischemia.

Methods: Male Sprague-Dawley rats were divided into five groups (n=24): sham, control ischemic, candesartan treated (0.3 mg/kg), alpha tocopherol treated (30 mg/kg) and combined treated ischemic groups. Transient focal cerebral ischemia was induced by 90-min-long occlusion of the left middle cerebral artery followed by 24-h-long reperfusion. Neurological deficit score was evaluated at the end of the reperfusion period. Thereafter, the animals were randomly used for measurement of the infarct volumes and investigation of ischemic brain edema formation using a wet/dry method.

Results: Induction of cerebral ischemia produced considerable brain infarction in conjunction with severely impaired motor functions and edema formation. Combined treatment with candesartan and alpha tocopherol significantly reduced the infarct volume and lowered the water content in the ischemic lesioned hemisphere. These effects on brain edema and oxidative stress biomarkers were significantly more than the monotherapy with candesartan.

Conclusion: The combination therapy with candesartan and alpha tocopherol can noticeably decrease ischemic brain injury and attenuate edema formation likely via increasing the antioxidant activity.

Abstract

Background & objectives: Cyclophosphamide is one of the common drugs used for cancer treatment. Cyclophosphamide affects the placenta and it is teratogenic in pregnant women. The purpose of this study was to evaluate the protective effects of Mesna and Galbanum essential oil concurrent administration against cyclophosphamide toxicity in rat placenta tissue.

Methods: 22 pregnant female rats were randomly divided into 3 groups: (1) control (normal saline, n=7 rats), (2) cyclophosphamide (15mg/kg, n=7 rats), (3) cyclophosphamide (15mg/kg), Mesna (100mg/kg) and Galbanum (200mg/kg, n=8 rats). Medications were administered intraperitoneally on the 13th day of pregnancy. Then, rats were sacrificed on the 20th day of pregnancy. The placenta was separated and fixed after macroscopic evaluation. 5 µm tissue sections were prepared, stained and studied by light microscopy.

Results: Cyclophosphamide reduced the placental weight, thickness and the length of the small and large diameters. Histologically, it reduced the thickness of the labyrinth and basal layers, caused necrosis and congestion in these layers, reduced giant cells numbers and caused their nuclear shrinkage (p<0.05). Mesna and Galbanum oil had no effect on placental morphometry rather than cyclophosphamide (p>0.05) but histologically it could increase the labyrinth and basal layers thickness and also the giant cells numbers (p<0.05). It could also prevent nuclear shrinkage, giant cells and spongiotrophoblasts necrosis in the basal layer and reduce congestion in the labyrinth layer (p<0.05).

Conclusion: Mesna and Galbanum oil preserved the placenta of pregnant rat against toxic effects of cyclophosphamide probably by its antioxidant features and free radicals deletion.

Abstract

Background & objectives: Streptococcus pneumoniae is  one of the major causes of vaccine - preventable diseases worldwide. Current pneumococcal vaccines consist of serotype specific capsular polysaccharide antigen and do not offer full clinical protection against pneumococcal diseases. Due to such limitations, a new generation of protein-based pneumococcal vaccines is being developed. The objective of this study was to determine the distribution of gens encoding five protein antigens including pneumococcal histidine triad D and E (phtD, phtE), rlr- regulated gene A (rrgA), Autolysin (lytA) and Pneumococcal surface protein C (pcpC) among pneumococcal isolates collected from nasopharyngeal specimens in healthy children.

Methods: A total of 43 pneumococcal isolates were collected from nasopharyngeal specimens of healthy children attending the kindergartens in Ardabil province. The strains were identified using optochin susceptibility and bile solubility testes and further confirmed by amplification of capsular polysaccharide A gene (cpsA). PCR was used for screening the presence of pcpC, phtD, phtE, rrgA and lytA genes.

Results: 81.4 % of isolates were found to contain at least one of the tested genes. lytA, pcpC, phtE, phtD and rrgA were detected in 70, 60, 39.5, 35 and 25.5 percent of isolates, respectively. The results showed that the genes were not distributed consistently among the isolates and for obtaining a full coverage pneumococcal vaccine, multiple choices of these antigens should be included.
