Abstract

Background & Objectives: Since the students majoring in different Fields of medicine are going to play key roles regarding Community health in the future, it seams necessary to characterize problems threatening their mental health. This study was performed in order to evaluate students’ mental health status in Ardabil University of Medical Sciences
Methods: In this descriptive-analytical study, 352 students were selected through census and were requested to fill out general health questionnaire (GHQ) with 28 questions as well as personal information questionnaire. The data were analyzed using descriptive statistics and chi- square.
Results: The findings indicate that students have problems in physical (1.7%), and social function (4.3%), anxiety and insomnia (2.3%) of them suffer from severe depression (2%). Also according to cut point (23) 22.7% students were suspected of mental disorders. In this research, there was no significant difference in mental disorders between male and female students, but there was a meaningful relationship between status with paternal educational degree and family size and mother's job (p< 0.05).
Conclusion: according to the findings, student's psychological evaluation and counseling service in different fields to promote student mental health, is recommended

Abstract

Background & objectives: Breast cancer is one of the most important cancers in women worldwide. Taxol as a chemotherapeutic agent, is used for treatment of breast cancer. The aim of this study was to investigate alterations in the expression of mir-1246 and mir-224 in four breast cancer cell lines after Taxol treatment with the goal of introducing them as a biochemical marker for determining response or resistance of breast cancer to the Taxol therapy.
Methods: In this in vitro study, four breast cancer cell lines including MCF-7, MDA-MB-231, SKBR-3 and BT-474 were cultured in RPMI1640 medium supplemented with 10% FBS and antibiotics. Then, MTT assay was performed to determine IC50 concentration of Taxol. Cells were treated for 24 hours and then RNA extraction and cDNA synthesis were performed. Alterations in the expression level of mir-1246 and mir-224 were quantitated using qRT- PCR.
Results: After treatment with Taxol, the expression level of mir-1246 was significantly up-regulated in two HER2-overexpressing cell lines, BT-474 (113 fold) and SKBR-3 (1.4 fold), and down-regulated in two HER2-negative cell lines, MCF-7 (45.5 fold) and MDA-MB-231 (7.7 fold). Expression of mir-224 was detected only in two cell lines including SKBR-3 and MDA-MB-231, and was down-regulated after treatment with Taxol (2.1 and 17.2 fold, respectively).
Conclusions: According to the different pattern of alteration in the expression level of mir-1246 in HER2-overexpressing breast cancer cell lines compared to HER2-negative cell lines after treatment with Taxol, this miRNA could be a useful biomarker for responsiveness to Taxol in  different types of HER2-positive and -negative breast cancers.

Abstract

Background & objectives: Vincristine (VIN) is a broad-spectrum anticancer drug has been used to treat various cancers. Resveratrol (Res) is a natural polyphenol found in many plant sources. Many studies have reported anti-inflammatory and antioxidative effects of resveratrol. We have explored the protective effect of resveratrol on vincristine-induced oxidative stress in mouse ovarian tissue.
Methods: In this study, 32 female mice weighing 25-30 grams were randomly divided into four groups (each group n=8): 1- control group, 2- Vin- group, 3- Vin-Res group and 4- Res group. The mice received a single IP injection of vincristine (3 mg/kg) weekly for 4 weeks. Res treatment was done 28 days by gastric gavage (daily 30 mg/kg).At the end of the study, Malondialdehyde (MDA) and total antioxidant capacity (TAC) and the antioxidant activity of superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzymes were measured in ovarian tissue and uterus of the studied animals. Also, ovarian follicles were counted.
Results: The results indicated that the MDA level was elevated and TAC, GPx as well as SOD activities were decreased in Vin- group significantly. Resveratrol reduced MDA level and increased GPx and SOD activities in Vin-Res group significantly. Also histological findings showed that Res increased primordial and primary follicles and reduced atretic  follicles in Vin-Res group significantly.
Conclusions: These results indicate the protective effect of resveratrol on ovarian and uterine tissue against oxidative damage of vincristine in mice
